私はしばらくの間読み回しましたが、これを機能させることができません。

mod_rewrite は Apache で有効になっており、AllowOverride を all に変更しました。

コードを次のように変更してみてください。

Options +FollowSymLinks -MultiViews
# Turn mod_rewrite on
RewriteEngine On
RewriteBase /

RewriteCond %{REQUEST_FILENAME} !-f
RewriteCond %{REQUEST_FILENAME} !-d
RewriteRule ^(.+)$ profile.php/?username=$1 [L]

つまり、MultiViewsオプションをオフにします。

PS: error.log を見ると、RewriteEngineキーワードの前に .htaccess に特殊文字が含まれているようです。
